Core Facility: Toxic Algal Culture Facility Principal Investigators: Kelly
S. Rein Ph.D. and Chris Sinigalliano
Ph.D. The Toxic Algal Culture Core Facility is located at Florida International University (FIU). This Facilities Core provides field and laboratory support to the Toxic HAB research project (Brand/Rein), and the HAB Functional Genomics research project (Crawford/Hitchcock) as well as other Oceans and Human Health investigators. The Culture Facility: (1) Collects and analyzes samples at sea via a continuous-flow bow pump system linked to a flow-through chlorophyll fluorometer and flow cytometer. (2) Isolates total population DNA from field collected samples for the preparation of 16S and 23S libraries to assess population diversity. (3) Isolates RNA from field-collected samples for studies of gene expression. (4) Establishes uni-algal and axenic (whenever possible) cultures of phytoplankton (dinoflagellates, prymnesiophytes and raphidophyts) by both traditional manual and automated cell-sorting (flow cytometry) methods. (5) Maintains clonal cultures and large-scale cultures. (6) Collects biomass from large-scale cultures. (7) Grows microorganisms under a variety of conditions for gene expression studies. Below is a list of dinoflagellate cultures that are now available.
